[Abstract]
Background and objective: Colorectal cancer is one of the most common malignancies in men and women. Despite recent advances in the treatment of colorectal cancer, the overall survival rate for those patients with advanced locoregional disease remains less than 50%. Although adjuvant systemic chemotherapy has improved survival of these patients, more effective therapies are needed. Tumor vaccine is an approach that could have a particular role in the adjuvant therapy of colorectal cancer. In this study, we designed a new efficient CEA vaccine of fussion epitopes with the vehicle of HPV16 L1, and expressed it in eukaryotic baculoviruse expression system.Methods: 1? The encoding fragment of HPV16 ? L1(34aa deleted in C terminal of HPV16 Ll)was amplified from the plasmid 114KL1pFastBacl by means of PCR and inserted into pMD18-T. 2? The fragment and CEA-PADRE synthesis primers was coloned into pFastBacl by restriction endonuclease and T4 ligase, formed a recombinant baculoviruse transfer vector of the vaccine. 3? The transfer vector was transposited with baculoviruse shuttle vector(bacmid)in the Max efficiency DHlOBac competent cells, constructed baculoviruse expression vector of the vaccine. 4? After transfection into sf9 cells, the recombinant baculoviruse was detected by PCR. 5? Immnohistochemical method was used to detect the expression of HPV16L1, p53 and PCNA in 45 patients with colorectal cacinoma.Results: 1? By the enzyme cut evaluation and sequencing, the recombinant baculoviruse transfer vector of the CEA vaccine. 2? The recombinant baculoviruse was identified by PCR. 3? The result for examination of 45 patients with colorectal carcinoma by immnohistochemical assay show:the positive rate of HPV16L1 is 46.7%(21/45),HPV16 infection present had no association with histopathologic type, histopathologic grade, clinical stage and sex; the positive rate of p53 is 53.3%(24/45), it has no association with HPV16 infection(P=0.286>0.05); the high positive rate of PCNA is 75.6%(34/45), it shows positive correlation with HPV16 infection(P= 0.0310.05).
